Start your computer in safe mode by pressing F8 Repeatedly during startup. It make look bad, but it starts using the minimum processes so the thing that is in use may not be in use.

I use this program called unlocker , when its installed you right click on item and youll see unlocker , click it and it will give you options and deal with it for you.
if u wanted to delete that file then i need to reboot the computer in safe mode and then goahead deleting.

press F8 when the system starts and get the advance boot menu to goto safemode.
zone alarm fire wall,spyware terminator and active virus shield is a perfect answer for you.
