You can also reinstall your operating system from scratch after formatting your hard drive, but you will lose all your data. The best thing to do is remove your HD, insert it into a USB or FireWire HD case, and plug it into an OS X or Linux computer. Copy your important data onto the OS X or Linux computer, put the HD back, and reinstall XP or Vista or whatever OS you were using on your PC. Then virus check your backed up data and copy it back to your PC if it is safe.

Some av websites, ie McAfee, CA, kaspersky, Symantec have free av scanners that you can use. Some also have free virus removal tools that you can use.
